Besides binding to host cell receptors, priming of the S glycoprotein by host proteases through cleavage at the S1/S2 and the S2′ sites is another crucial factor modulating tropism and pathogenicity. For instance, entry of the MERS-CoV-related bat coronavirus HKU4 into human cells required addition of exogenous trypsin, indicating that S proteolytic activation of this bat virus did not occur in human cells (despite its ability to recognize human DPP4).
