Despite their different tertiary structures, HCoV-NL63 and SARS-CoV S1-CTDs bind to a common region on ACE2. The VBMs for two viruses on ACE2 overlap, and a number of ACE2 residues interact with both S1-CTDs. Surprisingly, one of the two virus-binding hot spots on ACE2 for SARS-CoV binding, which centers on ACE2 residue Lys353, plays a similarly critical role in the binding of HCoV-NL63.
